Abstract
A successful peripheral nerve block requires knowledge of physiology and pharmacology of local anesthetics (LAs). This article explains pharmacodynamics of LAs, differential blockade, tachyphylaxis, C-fiber resistance, tonic/phasic blockade, and volume/concentration effects. New neurotoxin-based anesthetics show potential.
